Polyethylene glycol auxiliary application: pharmaceutical preparations
1 Pharmaceutical solvents
PEG200, PEG300, PEG400, PEG600 are colorless, slightly smelly viscous liquids with stable chemical properties and low toxicity, so they are often used as solvents for drugs. In addition, in order to increase the solubility of insoluble drugs, the latent solvent is often used, that is, ethanol, glycerol, propylene glycol, benzyl alcohol, polyethylene glycol and other mixed solvents composed of water.
2 Lubricants and adhesives
PEG4000 and PEG6000 are typical representatives of water-soluble lubricants in tablets. Appropriate amount of polyethylene glycol can be added directly into the tablet prescription for whole granulation, or it can be prepared into alcohol solution, suspension or emulsion for granulation, and the lubrication effect remains unchanged. The disintegration and dissolution of tablets prepared with polyethylene glycol are not affected, which can improve the solubility of the main drug in the stomach, and ultimately contribute to increased bioavailability.
3 Finishing material
Polyethylene glycol (PEG) modifiers are pH neutral, non-toxic, water-soluble polymers with high hydrophilicity and good biocompatibility and blood compatibility, and no immunogenicity. Therefore, structural modification with PEG can improve the following properties of drugs:
(1) Increase stability and reduce enzymatic degradation;
(2) Improving pharmacokinetic properties, such as prolonging plasma half-life, decreasing maximum blood concentration, decreasing blood concentration fluctuation, etc.;
(3) decreased immunogenicity and antigenicity;
(4) Reduce toxicity and improve in vivo activity;
(5) Improved drug distribution in the body, enhanced targeting;
(6) Reduce the frequency of medication and improve patient compliance.
